1-乙基-6-氟-1,4-二氢-4-氧代-7-(1-哌嗪基)-3-喹啉羧酸-铜(Ⅱ)-多吡啶配合物的合成、表征及与DNA作用

摘要: 合成了4个新的诺氟沙星(NFA)-铜(Ⅱ)-多吡啶 配合物:[Cu(NFA)(phen)(H2O)](ClO4)2 (1),[Cu(NFA)(tatp)(H2O)](ClO4)2 (2), [Cu(NFA)(dppz)(H2O)](ClO4)2·H2O (3)和[Cu(NFA)(n-phen)(H2O)](ClO4)2 (4)[NFA=1-乙基-6-氟-1,4-二氢-4-氧代-7-(1-哌嗪基)-3-喹啉羧酸,phen=1,10-邻菲咯啉,tatp=1,4,8,9-四氮三联苯,dppz=二吡啶并[3,2-a;2′,3′-c]吩嗪,n-phen=5-硝基-1,10-邻菲咯啉]。用元素分析、摩尔电导率、红外光谱及紫外可见光谱对配合物进行了表征,推测了配合物的可能结构。应用紫外光谱、荧光光谱及粘度测定方法研究了配合物与CT-DNA的作用,发现这些配合物均以插入方式与CT-DNA作用。采用打孔法研究了配合物的抗菌活性,结果表明配合物对耻垢分枝杆菌(M.smegmayis)有良好的抑制作用。

Abstract: Four new complexes: [Cu(NFA)(phen)(H2O)](ClO4)2 (1), [Cu(NFA)(tatp)(H2O)](ClO4)2 (2), [Cu(NFA)(dppz)(H2O)](ClO4)2·H2O (3) and [Cu(NFA)(n-phen) (H2O)](ClO4)2 (4) (NFA=1-Ethyl-6-fluoro-1,4-dihydro-4-oxo-7-(1-piperazinyl)-quinoline-3-carboxylic acid (Norfloxacin), phen=1,10-phenanthroline, tatp=1,4,8,9-tetraazatniphen-ylene, dppz=dipyrido[3,2-a; 2′,3′-c]phenazine, and n-phen=5-nitro-1,10-phenanthroline), were synthesized and character-ized by elemental analysis, molar conductivity, IR and UV-Vis spectroscopes. The interaction between the complexes and CT-DNA was also studied by electronic absorption spectroscopy, ethidium bromide(EB) fluorescence spectroscopy, and viscosity measurements. In addition, the complexes were assayed against M.smegmayis by punch method.
