Abstract: The fluorescence quenching spectra of the holo-CuA and apo-CuA domain of cytochrome c oxidase from Paracoccus versutus by KI were investigated.The results indicated that in solution part of the fluorophore tryptophanes of the CuA domain are exposed to the surface of the protein,and others buried inside of the hydrophobic core.The fluorescence quenching constants of holo-CuA and apo-CuA domain protein by KI are 1.39 and 2.24,respectively,which suggested that the bi-nuclear copper center [CuA+1.5-CuA+1.5] also plays a role on the structural stability of the protein.
