摘要: 用溶胶-凝胶制备了多孔透明的二氧化硅包埋肌红蛋白,在还原辅酶Ⅰ二钠盐(NADH)和黄素单核苷酸(FMN)的存在下,用可见光谱法测定了肌红蛋白苯胺羟基化反应的Km和kcat。表明包埋在二氧化硅中的肌红蛋白保持了苯胺羟化酶活性。 |

Abstract: Using sol-gel procedures, we encapsulated myoglobin in the porous and transparent silicate glass. The transparency of sol-gel makes it feasible for us to monitor the hydroxylation reaction spectroscopically in the pore of the matrix. Thus the Km and kcat values of the hydroxylation reaction for Mb-sol-gel were determined to be 10.3 mmol·L-1 and 0.013 min-1, respectively. It shows that the aniline-hydroxylase activity of myoglobin (Mb) encapsulated in silicte sol-gels in maintained and can be induced by a semi-artificial multienzyme system of NADH-FMN-Mb. |
