牛血清白蛋白与四氯合铂(Ⅱ)酸根、顺铂及顺式二水二氨合铂(Ⅱ)相互作用的研究
INTERACTIONS BETWEEN BOVINE SERUM ALBUMIN (BSA) WITH TETRACHLOROPLATINATE(II), CISPLATIN AND CIS-DIAQUODIAMMINEPLATIN(II)

摘要: 用平衡透析、凝胶色谱等方法研究了牛血清白蛋白(BSA)与PtCl42-、顺铂及cis-[Pt(NH3)2(H2O)2]2+的相互作用。结果表明,顺铂与BSA在25℃作用4小时,未发现BSA结合铂,而在37℃作用20小时后BSA与铂(Ⅱ)有一定的结合。BSA对PtCl42-有一个强结合部位,3个弱结合部位,结合常数分别为4.0×104及1.1×103。BSA对cis-[Pt(NH3)2(H2O)2]2+有2个专一性结合部位,结合常数K1=9.0×104,有2-3个非专一性结合部位,K2=2.4×102,BSA与cis-[Pt(NH3)2(H2O)2]2+的反应对BSA是一级反应,速度常数k=0.012分-1

Abstract: The interactions between BSA and PtCl42-, cisplatin and cis-diaquodiammineplatin(II) have been studied by gel chromatography and equilibrium dialysis. The experimental results show that no platinization of BSA has been observed up to 4 hrs. at 25℃. However, after 20 hrs of reaction a small fraction of BSA has been platinized at 37℃. It is found that BSA has two groups of binding sites to PtCl42-, which has been characterized with number of binding sites 1 and 3, the binding constant are 4.0×104 and 1.1×103, respectively. For binding of cis-[Pt(NH3)2(H2O)2]2+, the number of sites in the first specific group equals to 2, the binding constant, 9.0 × 104 and the second, nonspecific, consists of 2-3 binding sites, binding constant is 2.4×103. The reaction between BSA and diaquo complex has been characterized as a first order reaction with k=0.012mm-1 at 37℃.
